Is it possible the second distance to be marked directly in points in the screen?

The short answer is no.

Since the first time I used this, I thought that it would be intuitive to do it that way. Instead, I start the Enlarge command, select CalDist and then realize that I do not have the target value, so, I quit the command, measure the target distance, write it down, then restart the command and input the target as the last value.

So, you have to know the value before you start.
